Functional Performance

General workflow:

  • Thaw at 37°C (~3 minutes)
  • Remove cryoprotectant with centrifugation
  • Resuspend in an appropriate activation buffer/normal plasma
  • Stimulate with agonists
  • Measure functional readouts

Granule Release Assays

Retham™ Cryoplatelets demonstrate functional responses in granule release formats, including:

  • Responsiveness to TRAP (Thrombin receptor activating peptide)
  • Intact FcGRIIa (CD32)-mediated signaling as evidenced by antibody-mediated activation

Aggregation-Based Assays

Demonstrated responsiveness to multiple agonists:

  • TRAP
  • Collagen
  • Ristocetin
  • ADP
  • Epinephrine
  • Arachidonic Acid

While higher agonist concentrations may be required versus fresh platelets, Retham™ Cryoplatelets offer broad agonist responsiveness, unlike many lyophilized alternatives.

Procoagulant Activity

Retham™ Cryoplatelets support robust coagulation-based assays:

  • Thrombin Generation (CAT): ~2x higher thrombin generation vs. fresh platelets
  • Thromboelastography: Strong clot formation
  • Shortened lag phase observed across assays

These characteristics suggest a procoagulant phenotype, potentially reflecting partial activation or altered membrane dynamics post-cryopreservation.
